What it is
Chemically, magnesium bisglycinate is one magnesium ion held between two glycine molecules — a structure called a chelate, from the Greek for “claw”. The glycine wraps around the magnesium and shields it, so the mineral stays dissolved through the changing acidity of the digestive tract instead of falling out of solution the way inorganic salts can. Glycine is the smallest amino acid, found in protein-rich foods and made by the body itself; it doubles as a calming signalling molecule in the nervous system.
The salt is about 14% elemental magnesium — so 100 mg of magnesium bisglycinate provides roughly 14 mg of actual magnesium. One label trap is worth knowing: some “buffered” bisglycinate products are blended with cheaper magnesium oxide to bump up the elemental figure. If a label promises an unusually high magnesium content for a glycinate product, that is usually why.
Absorption & bioavailability
Glycinate’s reputation rests on two properties. First, the chelate stays soluble along the whole gut, so more of it is available to absorb. Second, part of the dose appears to be taken up through peptide absorption routes — the channels the body uses for protein fragments — rather than competing only for the ordinary mineral transporters. The clearest human demonstration comes from patients whose small intestine had been surgically shortened: magnesium diglycinate was absorbed better than magnesium oxide precisely where ordinary mineral absorption is most compromised.1
The practical consequence of good absorption is comfort: little magnesium is left behind in the gut to draw in water, which is why glycinate is the form least likely to cause loose stools. What it’s used for
The benefits of glycinate are, first and foremost, the benefits of magnesium itself: correcting a low intake, and the muscle, sleep, mood, blood-pressure and migraine effects covered in the main guide. Glycine acts as a calming neurotransmitter, which is the rationale for reaching for glycinate specifically around sleep, stress and evening wind-down.
Honest footnote: the sleep evidence is for magnesium in general, not this form in particular. A meta-analysis of trials in older adults with insomnia found magnesium shortened time-to-sleep modestly, with the authors rating the overall evidence quality as low.2 So think of glycinate as the sensible way to take magnesium if sleep is your goal — not as a sleeping pill.
Dose & timing
The usual supplemental range is 200–400 mg elemental magnesium per day, taken with food to improve uptake and comfort. Doses always refer to elemental magnesium — a capsule labelled “1000 mg magnesium bisglycinate” contains roughly 140 mg of the mineral itself. Many people take it in the evening for the calming association; split morning/evening doses absorb marginally better if you are at the higher end. Side effects & safety
At normal doses glycinate is very well tolerated — loose stools are uncommon compared with other forms.
